If you play lots and lots you will develop a nice big callous. If it is happenening while chording, not to worry. If while playing single string, you might want to have a look at hand position in a mirror and see if you can figure out why.

I concur Jazzaferri's comments, I often have a black mark on my index from chording but single string playing should be clean. If you have an iPad or what not, film your hand position when playing and check it out against players like Rosenberg or these 3
